Comparison of Sequencing the same plasmid by two different methods: directly vs. prior amplification with Templiphi
 

Below are electropherograms showing sequence from the same plasmid prepared two different ways. In the lower panel, the plasmid (solid phase extraction kit) was sequenced directly with the standard sequencing protocol and has significant slippage due to secondary structure. In the upper panel, the sequencing templete was generated with an illustra Templiphi Sequence Resolver reaction from the same plasmid as below and the slippage has been completely eliminated.
