Comparison of Sequencing the same plasmid by two different methods: directly vs. amplification with Templiphi
 

Below are electropherograms showing sequence from the same plasmid prepared two different ways. In the lower panel, the plasmid was sequenced directly with the standard sequencing protocol and has significant slippage due to secondary structure. In the upper panel, the templete was generated with a Templiphi reaction and the slippage has been completely resolved. These results are courtesy of Assaf Katz Zondek and Michael Ibba.
